Nicotinamide mononucleotide, commonly abbreviated NMN, is a naturally occurring nucleotide found in the cells of many organisms. Its structure consists of a nicotinamide group linked to a ribose sugar that carries a phosphate group. NMN is an intermediate in the biosynthesis of nicotinamide adenine dinucleotide, or NAD+, a coenzyme involved in many metabolic reactions. The abbreviation usually refers to the beta anomer, though related forms can exist. In scientific literature, NMN is distinct from nicotinamide riboside, another NAD+ precursor.
In the NAD+ salvage pathway, the enzyme NAMPT converts nicotinamide and a phosphate-donor molecule into NMN. A second enzyme, NMNAT, then converts NMN into NAD+. Nicotinamide riboside can also enter this route after being converted to NMN by nicotinamide riboside kinases. Because NMN sits at a junction between precursor uptake and NAD+ formation, its cellular concentration is tightly linked to enzyme activity and tissue type. NAD+ participates in redox reactions, signaling, and DNA repair, and its levels decline with age in some animal models, though human evidence remains more limited and context-dependent.
Research interest in NMN increased after animal studies reported that oral or injected NMN can raise NAD+ levels in some tissues. How NMN is absorbed and distributed in humans is not fully established. Some evidence suggests extracellular NMN may be dephosphorylated to nicotinamide riboside before cellular uptake, while other studies propose specific transport routes. Direct human data on these mechanisms remain limited. Regulatory status also varies: in some countries NMN is treated as a dietary supplement, while elsewhere it is restricted or requires approval, and these differences affect labeling, sale, and research.
Stability testing examines how temperature, humidity, light, and pH affect NMN over time. The compound is generally stored cold, often at minus twenty degrees Celsius or lower, in a desiccated container protected from light. Aqueous solutions tend to be less stable than dry powder because hydrolysis and dephosphorylation can occur, potentially forming nicotinamide riboside or other degradation products. Stress studies may expose samples to heat, acid, base, oxidation, and strong light to identify likely degradation pathways. Results from such studies help define shelf life and handling recommendations, though exact stability depends on formulation and packaging.
Quality control for NMN materials typically includes identity, assay, purity, and impurity profiling. Tests may cover residual solvents, heavy metals, microbial limits, and water content, depending on the intended use and local rules. Impurity profiles can include related substances such as nicotinamide, nicotinamide riboside, and NAD+, which may form during synthesis or storage. Because commercial NMN can be offered as different salts or hydrate forms, a certificate of analysis should state the form and the analytical methods used. Independent verification is relevant because supplement markets vary in testing requirements and enforcement.
Laboratory identification of NMN usually relies on chromatographic separation coupled with ultraviolet or mass spectrometric detection. High-performance liquid chromatography with UV absorbance can quantify the compound against a reference standard, while liquid chromatography-tandem mass spectrometry offers lower detection limits and better specificity in complex matrices. Nuclear magnetic resonance spectroscopy can confirm structural identity and isomeric form. Ion chromatography or capillary electrophoresis may be used to identify counterions such as sodium. Method validation includes accuracy, precision, linearity, and limits of detection.

NMN occurs in many living systems, including bacteria, yeast, plants, and mammals. Dietary sources are present in foods such as edamame, avocado, broccoli, and various meats, but amounts vary widely and are generally lower than those used in research settings. Laboratory production often relies on enzymatic synthesis or chemical phosphorylation of nicotinamide riboside, and commercial material is typically supplied as a white to off-white powder. Because NMN is hygroscopic and sensitive to heat, moisture, and pH extremes, its handling requires care to preserve identity and purity. Aqueous preparation should be done with attention to pH and temperature to limit hydrolysis.
Nicotinamide mononucleotide, abbreviated NMN, is a naturally occurring nucleotide found in cells. Its structure consists of a nicotinamide ring linked to ribose phosphate, and the compound serves as an intermediate in the salvage pathway for nicotinamide adenine dinucleotide, or NAD+. In this pathway, nicotinamide phosphoribosyltransferase converts nicotinamide and phosphoribosyl pyrophosphate into NMN, after which NMN adenylyltransferase attaches an adenylate group to produce NAD+. Because NAD+ participates in redox reactions and signaling, NMN occupies a central position in cellular metabolism. The molecule is distinct from nicotinamide riboside, though the two are related in NAD+ precursor research.
Beyond its intracellular synthesis, NMN can be taken up from the extracellular environment, although the routes are still debated. Some evidence points to direct transport into cells through specific transporters, while other work suggests dephosphorylation to nicotinamide riboside followed by cellular uptake. Once inside, NMN can be converted to NAD+ by NMN adenylyltransferases; the relative contribution of these routes may differ by tissue, species, and experimental conditions. Researchers continue to investigate which mechanisms dominate in intact organisms and how they affect measured NAD+ levels. Direct measurement in tissues remains technically challenging because NMN can be rapidly metabolized during sample collection.
Regulatory treatment varies by country. In the United States, NMN has been marketed as a dietary supplement, but the Food and Drug Administration has stated that it is excluded from the dietary supplement definition because it was authorized for investigation as a new drug before being marketed as a supplement. Other jurisdictions may treat it as a novel food, a supplement, or an unapproved drug ingredient. Import and sale rules can therefore differ substantially.
Quality control for NMN focuses on identity, purity, residual solvents, heavy metals, and microbial limits. Because the molecule can absorb water, moisture content and packaging are relevant to shelf life. Suppliers may provide certificates of analysis, but independent verification is often needed for research or commercial use. The long-term stability of different crystal forms, salt forms, and formulations is not fully characterized in the public literature. Some degradation products and their effects on product performance remain open questions.

==== Droplet sorting ==== Droplet sorting in microfluidics is an important technique, allowing for discrimination based on factors ranging from droplet size to chemicals labeled with fluorescent tags within the droplet, stemming off of the work done to sort cells in Flow Cytometry. Within the realm of droplet sorting there are two main types, bulk sorting, which uses either active or passive methods, and precise sorting, which relies mainly on active methods. Bulk sorting is applied to samples with a large number of droplets (> 2000 s−1) that can be sorted based on intrinsic properties of the droplets (such as viscosity, density, etc.) without checking each droplet. Precise sorting, on the other hand, aims to separate droplets that meet certain criteria that is checked on each droplet. Passive sorting is done through control of the microfluidic channel design, allowing for discrimination based on droplet size. Size sorting relies on the bifurcating junctions in the channel to divert the flow, which causes droplets to sort based on how they interact with the cross section of that flow, the shear rate, which relates directly to their size. Other passive methods include inertia and microfiltration, each having to do with the physical properties, such as inertia, and density, of the droplet. Active sorting uses additional devices attached to the microfluidic device to alter the path of a droplet during flow by controlling some aspect, including thermal, magnetic, pneumatic, acoustic, hydrodynamic and electric control.
== History == Somapacitan was evaluated in a randomized, double-blind, placebo-controlled trial (NCT02229851) in 300 participants with growth hormone deficiency who had never received growth hormone treatment or had stopped treatment with other growth hormone formulations at least three months before the study. Participants were randomly assigned to receive injections of weekly somapacitan, weekly placebo (inactive treatment), or daily somatropin, an FDA-approved growth hormone. The effectiveness of somapacitan was determined by the percentage change of truncal fat, the fat that is accumulated in the trunk or central area of the body that is regulated by growth hormone and can be associated with serious medical issues. The trial was conducted at 92 sites in 16 countries: the United States, Australia, Germany, India, Japan, Latvia, Lithuania, Malaysia, Poland, Romania, Russian Fed, South Africa, Sweden, Turkey, Ukraine and the United Kingdom. Adult participants were assigned at random to weekly Sogroya or placebo injections for 34 weeks. Neither the participants nor the investigators knew which treatment was given until the end of the trial. One additional group of participants with GHD received daily injections of somatotropin (an approved treatment for GHD). At the end of the 34-week treatment period, truncal fat decreased by 1.06%, on average, among participants taking weekly somapacitan while it increased among participants taking the placebo by 0.47%. In the daily somatropin group, truncal fat decreased by 2.23%.
The RF's intention was partly to test whether or not the British would attempt to block this bill after Gibbs had granted Royal Assent to it, but this issue never came to a head because Sandys persuaded Field not to forward it to Gibbs for ratification on the grounds that it had not been unanimously passed. Lord Salisbury, one of Southern Rhodesia's main supporters in Britain, despaired at Field's lack of action, telling Welensky that as he saw it "the simple time to have declared independence, whether right or wrong, would have been when the Federation came to an end". The RF hierarchy interpreted this latest backtrack by Field as evidence that he would not seriously challenge the British on the independence issue, and forced his resignation on 13 April 1964. Smith accepted the Cabinet's nomination to take his place.

Diphenhydramine is not typically used to treat anxiety because its long-term use may cause adverse effects, such as memory loss, especially in the elderly. Diphenhydramine is not approved by the US Food and Drug Administration (FDA) for treating anxiety. On the other hand, hydroxyzine, a first-generation antihistamine that lacks significant anticholinergic effects, may be used to treat anxiety, although benzodiazepines and antidepressants are considered more effective by most clinicians. The mild anxiolytic effects of hydroxyzine are mostly due to its weak but significant activity as an antagonist of the 5-HT2A receptor, a common target of most antidepressant drugs (as well as certain other antihistamines like cyproheptadine and promethazine). Diphenhydramine is not known to bind to the 5-HT2A receptor, though it is a weak antagonist of the related 5-HT2C receptor, which is another target of antidepressant drugs and has a significant role in mood and anxiety.
12 October Anti-war protesters invaded a U.S. Army base for the first time, as an estimated 5,000 anti-war demonstrators crossed into the boundaries of the base at Fort Dix, New Jersey. The group was driven back by about 1,000 military policemen with tear gas, and there were no arrests and no injuries.
=== Biomedical applications === Ethyl carbamate was originally used as an antineoplastic agent, hypnotic, sclerosing agent, adjunct to sulfonamide therapy, and topical bactericide. It was discovered to be carcinogenic in 1943 and largely ineffective at treating multiple myeloma, according to a study published in 1966. It was removed from the market in the US, UK and Canada in 1963 and was classified as a 2B carcinogen by the IARC in 1974. However, its use in Japan as a co-solvent in water for dissolving water-insoluble analgesics used for post-operation pain, at doses estimated to be at levels that are carcinogenic in mice, continued until 1975. Miller has suggested that this may have been the "largest number (millions) of humans exposed to the largest doses of a pure carcinogen that is on record" and called for studies to determine the effects on Japanese cancer rates to be performed. Ethyl carbamate continues to be used as an anaesthetic in animal experiments, with more than 100 animal studies using ethyl carbamate published each year. One advantage of using ethyl carbamate is that it has a very long duration of action, with some adult rats remaining anaesthetised 24 hours after administration of the drug. It also does not depress neuronal activity in the cortex to the same extent as isoflurane.
Tetra Brik is a brand name for a carton package produced by the Swedish packaging company Tetra Pak. Its shape is cubic or cuboid, and it is available with or without various different caps. The Tetra Brik is the best-known and most sold package in the Tetra Pak packaging family, to the extent that Tetra Brik is sometimes mistaken for the brand name. The Tetra Brik comes in either chilled (Tetra Brik) or ambient (Tetra Brik Aseptic) package types.
=== Immune system === Strong statistical evidence links vitiligo to changes in the immune system. It is thought to be caused by the immune system attacking and destroying melanocytes. Variations in genes expressed in immune cells or melanocytes have been associated with the disorder. A genome-wide association study found approximately 36 independent susceptibility loci for generalized vitiligo. One of them is the gene that encodes the protein tyrosinase, a melanocyte enzyme that catalyzes melanin biosynthesis and is a major autoantigen in generalized vitiligo. It has been hypothesized that damaging environmental factors can disrupt redox reactions necessary for protein folding, so skin cells may initiate the unfolded protein response, which releases cytokines and thus triggers an immune response. Additionally, artificial sweeteners such as sucralose can make gut bacteria more aggressive, potentially damaging pigment-producing cells. Vitiligo is sometimes associated with autoimmune and inflammatory diseases such as Hashimoto's thyroiditis, scleroderma, rheumatoid arthritis, type 1 diabetes mellitus, psoriasis, Addison's disease, pernicious anemia, alopecia areata, systemic lupus erythematosus, and celiac disease. Among the inflammatory products of NLRP1 are caspase 1 and caspase 7, which activate the inflammatory cytokine interleukin-1β. Interleukin-1β and interleukin-18 are expressed at high levels in people with vitiligo. In one of the mutations, the amino acid leucine in the NALP1 protein was replaced by histidine (Leu155 → His).

Nicotinamide mononucleotide is a nucleotide intermediate in the biosynthesis of NAD+. It consists of nicotinamide attached to a ribose phosphate unit. NMN occurs naturally in cells and is present at low levels in some foods.
NMN is a direct precursor in the NAD+ salvage pathway. Enzymes called NMNAT convert NMN into NAD+, a coenzyme used in metabolism and cell signaling. Raising NMN may increase NAD+ in some experimental settings, but the effect depends on tissue and organism.
No. Nicotinamide riboside is a related compound that lacks the phosphate group present in NMN. Cells can convert nicotinamide riboside into NMN, and both compounds feed into NAD+ production through overlapping routes.
NMN is often measured by high-performance liquid chromatography with ultraviolet detection. Liquid chromatography-tandem mass spectrometry can provide greater sensitivity and specificity. The chosen method should be validated and compared against a certified reference standard when possible.
